Design Notes

The trans element is an activating riboregulator which binds to a complimentary region on the cis repressor and opens up the RBS of the previously-repressed gene. When a plasmid containing this construct is present in a system that has the appropriate cis-repressed gene, activation of that gene ensues. Trans4 activates cis7 Design specifics: free energy = -24.7kcal; 4bp loop, 3 bulges, 16bp tail

The pBad and TT portions of the sequence came from the pCS958 template from the Smolke lab. The ta region was designed as an insert between these regions.
